I’d need to know more, but if it’s training the immune system not to overreact to the presence of an allergen, I suspect they’re targeting specific protein strands that keep mast cells stable and from degranulating–which is what happens when you’re exposed to a trigger.

So, yes, this could be significant in terms of calming down and reducing the over-priming of mast cells that triggers both normal and idiopathic reactions we see in mast cell diseases.

And even if it doesn’t work (though I can’t think of a reason why it wouldn’t) this is still incredible. Holy shit, what a time to be alive.

I think it is slightly more complex than that (but be advised that I did not take immunology and am relying on Claude to explain some of the terms).

The vaccine seems to be the simplest possible implementation: it’s just the code for the protein you’re allergic to. Allergic to a peanut protein? We’re gonna tell your cells to make that exact peanut protein.

That seems like it should just give you an allergy attack!

Except that immune systems are super complicated. This is where I had to ask Claude; there are a bunch of different kinds of T cells, and apparently having your own cells producing peanut protein (or whatever) into your bloodstream causes a different family of T cells to activate than activates when the protein is encountered in your airways or food.

And that activation shift is so strong that even if you later encounter the problematic protein in your airways or food, your immune system does not care and does not shift over to the types of T cells that tell your mast cells to freak out and dump histamine.

And they showed that they can make the mice allergic first and vaccinate them after, it’s the last experiment they lost, and it works fine.

As far as I can tell they are basically tricking your immune system into searching for the thing you’re allergic to in your bloodstream and on the surfaces of your cells instead of looking for it showing up in your airways or digestive tract. This may have side effects, as you’re ramping up the T cells that kill your own infected cells, but since you’re not actually infected with a virus that is making the protein in question, this should not be a problem.

Many things that work in mouse models don’t translate over to human treatments, so I would not be more than cautiously optimistic.

However, we know that allergy shots work for many people, and this is basically a time release allergy shot. Which should be safer and probably work better.

It is specifically Th2 responses that are getting down regulated, for anyone trying to dig deeper.

Ah, I was kinda right. So they’re trying to modulate the Th2 Cytokine response, which is what causes mast cells to degranulate and cause allergic reactions.

This specific approach may not help those of us with purely idiopathic anaphylaxis (meaning we test negative for our allergens because we’re missing the IgE protein that most people test positive for when they have allergic reactions. It’s a different thing we’re reacting to because mast cells have a ton of different mediators and it’s the mast cells themselves which are broke in our case), but I do wonder if it could be done in such a way to lower the overall allergic inflammatory response by getting the Th2 cells to chill out and not send out false signals.
